Résumé

Les auteurs quantifient et comparent les six performances (résolution, profondeur, sensibilité, portabilité, quantification, coût) des quatre techniques d’imagerie à l’échelle moléculaire (IRM, optique, ultrasons et PET), en correspondance avec les trois échelles du rayonnement électromagnétique utilisé in vivo (fréquences (106 à 1022 Hz), énergie photonique (10-4à 109 eV) et longueur d’onde (10-2à 10-15m). Cette imagerie moléculaire exige les technique les plus sensibles (Pl. 2-2. à 2-4). Quatre exemples d’approches expérimentales chez le petit animal, in vivo , sont montrés ici. Le passage moléculaire à travers la barrière hémato-encéphalique (cellules endothéliales, astrocytes et occludine, pharmaco-cinétique étudiée en TEP) (Pl. 2-5. à 2-11), d’abord, l’imagerie des récepteurs et des ligands notemment des récepteurs périphériques aux Benzodiazépines (PBR) en PET et IRM (Pl. 2-12 à Pl. 2-15), ensuite, la neuropathologie des maladies neuro-dégénératives et inflammatoires comme des accidents vasculaires cérébraux (PET et IRM chez le rat) (Pl. 2-16 et 17), et, enfin, l’étude de la réponses aux stimulations par l’imagerie in vivo des signaux calciques et de ses variations par l’analyse photonique, comme à l’échelle du Calcium mitochondria (Pl. 2-18 à Pl. 2-22).

Summary

We quantified and compared six parameters (resolution, depth, sensitivity, portability, quantification and cost) of four molecular imaging techniques (MRI, optics, ultrasound and TEP), with the three types of electromagnetic radiation used in vivo (Frequencies (106 to 1022 Hz), Photonic Energy (10-4 to 109 eV) and Wavelengths (10-2 to 10-15 m). This form of molecular imaging demands the most sensitive technique available (Pl. 26-2 to 26-4). Four examples of experimental in vivo approaches on small animals are shown: molecular passage through the blood-brain barrier (endothelial cells, astrocytes and occludin, pharmacokinetics, studied with PET) (Pl. 2-5. to 2-11) ; imaging of receptors and ligands, especially peripheral benzodiazepine receptors (PBR) by PET and MRI in the rat (Pl. 2-12 to Pl. 2-15) ; neuropathology of neurodegenerative and inflammatory diseases and stroke by PET and MRI in the rat (Pl. 2-16 to 2- 17) ; and the study of responses to stimulation explored with in vivo imaging of calcium signals and their variations by photonic analysis, as on the scale of mitochondrial calcium (Pl.2-18 to Pl.2-22).
